Huang Receives NIH Trailblazer Award to Advance Noninvasive Brain Drug Delivery

Zhimin Huang, PhDPittsburgh, August 4, 2026 — Dr. Zhimin Huang, Assistant Professor in the Department of Neurological Surgery, has been awarded an NIH/NIBIB Trailblazer Award to develop a next-generation platform for noninvasive, programmable drug delivery to the brain.

The three-year research project addresses one of the greatest challenges in treating neurological disorders: delivering therapeutic molecules precisely to diseased regions of the brain while minimizing effects on healthy tissue. Dr. Huang's team will combine engineered programmable exosomes, naturally occurring nanoscale vesicles that transport biological molecules between cells, with focused ultrasound, a noninvasive technology capable of submillimeter-precision targeting, to enable highly localized and programmable therapeutic delivery.

The new platform employs a molecular "masking" strategy to prevent engineered exosomes from interacting with cells outside the intended treatment site. Focused ultrasound is then applied to selectively "unmask" the exosomes only at the targeted location, enabling localized activation and site-specific delivery of therapeutic cargo without disrupting the blood-brain barrier. By combining molecular engineering with noninvasive ultrasound guidance, this approach enables spatiotemporally precise and repeatable drug delivery while minimizing off-target effects.

Beyond neurological disorders, this technology has the potential to benefit a wide range of diseases that require chronic, localized therapy. By integrating advances in bioengineering, synthetic biology, and focused ultrasound, the project aims to establish a versatile platform for the next generation of targeted therapeutics.
